This Human Semaphorin 4G ELISA Kit is intended for quantitative detection of human SEMA4G in cell culture supernates, serum and plasma (heparin, EDTA, citrate). Strip well format. Reagents for up to 96 tests.
This human SEMA4G ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for SEMA4G has been precoated onto 96-well plates. Standards (Expression system for standard: NSO, Immunogen sequence: V18-L675) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for SEMA4G is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human SEMA4G amount of sample captured in plate.

Expression system for standard: Semaphorin-4G is a protein that in humans is encoded by the SEMA4G gene. This gene is mapped to 10q24.31. Semaphorins are a large family of conserved secreted and membrane associated proteins which possess a semaphorin (Sema) domain and a PSI domain (found in plexins, semaphorins and integrins) in the N-terminal extracellular portion. Based on sequence and structural similarities, semaphorins are put into eight classes: invertebrates contain classes 1 and 2, viruses have class V, and vertebrates contain classes 3-7. Semaphorins serve as axon guidance ligands via multimeric receptor complexes, some (if not all) containing plexin proteins.
